Services/Core Grant

The following information is for investigators using the TAF and details what is expected of investigators as well as what to expect from the facility:

  1. Pre-Submission of order
  2. Submission of order
  3. Before microinjection
  4. After microinjection

Before submitting a Transgenic order, investigators need to prepare the following:

  1. Obtain a University approved animal protocol

  2. For first time users, you need to inform facility staff that you intend to use transgenic services.  TAF will file an Amendment Request to ILACUC and register you as a co-investigator on transgenic protocol #99A0010.  This procedure may take months to complete, so please initiate as early as possible.

  3. Once you are listed as a co-investigator on transgenic protocol #99A0010, you will be reminded to notifiy OSURF that funds from any relevant grants may be used to purchase animals on this protocol for the generation of your transgenic/knock-out animals.  This notification will allow OSURF personnel to verify that the TAF is permitted to use the account numbers you provide to order animals on your behalf.

  4. Work out a protocol to demonstrate the laboratory is able to detect a single copy of the transgene when it is mixed with tail DNA, or that the laboratory is capable of detecting an endogenous, single-copy, mouse gene (please see transgenic protocols).

When submitting a Transgenic order, investigators need to bring the following:

  1. Transgenic, RederivationBlastocyst Injection, Embryo Cryopreservation or Sperm Cryopreservation Service Order Form

  2. Completely digested DNA fragment to be injected (minimal 50µg - the more the better)

  3. Documentation of single copy detection of transgene

  4. Transgenic Facility/PI contract (please contact Xin-An for contract)

  5. Signed 100W

  6. Animal Requisition Form

Before microinjection of the DNA:

  1. TAF will purify the DNA fragment.  This process may be repeated when the quantity or quality of the DNA does not meet the desired standard.

  2. TAF will order in animals according to the scheduling.

After microinjection of the DNA:

  1. Investigators will be billed.

  2. TAF will inform investigators as soon as potential transgenic pups are born.

  3. The tails will be given to investigators two weeks after the pups are born.

  4. At weaning, potential transgenic pups will be shifted to PI's account.

  5. Transgenic founders will be transferred to PI's designated facility as soon as the genotyping result is given to TAF.

  6. TAF guarantees three transgenic founders for non-lethal constructs injected into FVB embryos, or one transgenic founder for non-lethal constructs injected into C57BL/6 embryos.
