Standard Pronuclear Microinjections
- $2500.00 per construct plus animal procurement and per diem
- TAF will microinject the transgene of interest into inbred FVB/N mice, and guarantee minimal three founders when the DNA fragment is purified by TAF for microinjection
- TAF will microinject the transgene of interest into inbred C57BL/6 mice, and guarantee minimal one founder when the DNA fragment is purified by TAF for microinjection
- Above guarantee of founders applies ONLY when Investigators provide documentation that a single copy of transgene is detectable by PCR or Southern when the transgene is diluted in mouse tail DNA
- Without documentation of detectable single copy of transgene, TAF will guarantee to inject minimal 250 zygotes or produce minimal 50 pups (whichever comes first). However, the founder guarantee does not apply.
YAC/BAC Transgenesis
- $2500.00 per construct plus animal procurement and per diem
- Investigators prepare the DNA for this procedure
- TAF will microinject the transgene of interest into minimal 200 FVB/N embryos
- The founder guarantee does not apply, although the TAF has been successful in all trials
Lentivial Transgenesis

- TAF will microinject the transgene of interest carried by lentivial vector into minimal 200 FVB/N embryos or the strain specified by PI

Blastocyst Microinjection
- $2500.00 per construct (up to 4 clones) plus animal procurement and per diem
- Investigators prepare the ES cells for microinjection
- TAF will microinject the ES cells into C57BL/6 blastocysts
- TAF will guarantee to inject up to 50 blastocysts per ES clone. However, germline transmission can not be guaranteed.

- If females of the desired strain are commercially available, the TAF will order the animal on behalf of investigators based on the schedule for the procedure

Cryopreservation Rational: Cryopreservation safeguards mouse strains against breeding failures, pathogens, and genetic contamination. Once a strain is cryo-preserved, un-needed live stocks of mouse can be eliminated from the colony. TAF will cryopreserve minimal 250 embryos with the desired genotype. Speed Cryo relies on in vitro fertilization (IVF) to generate 2-cell embryos for cryopreservation.
I. Freezing mouse sperms
- Fee: $350.00 per mouse line
- Investigators provide minimal 1-2, preferably 3-5, fertile stud male mice of desired genotype
- TAF will cryo-preserve minimal 4 vials of mouse sperms per line
- TAF will thaw one vial of frozen sperms and test the sperm motility
II. Freezing mouse embryos at 8-cell stage
II-1. Fee:
- $400-$650.00 per session plus the animal procurement and per diem (It usually takes three or more sessions varied by mouse lines.)
II-2. What needed from Investigators
- Cryopreservation Service Requisition Form
- When the mouse line is homozygous, PI provide both male and female mice
- Up to 15 fertile stud males that carry the desired modification with good breeding record
- Minimal 30, up to 45 female mice at 3-4 wk-old
- When the mouse line is heterozygous, and the female mice of the desired strain are commercially available, PI provide:
- Up to 15 fertile stud males that carry the desired modification with good breeding record
- Animal Requisition Form with billing information. TAF will order in juvenile female mice on behalf of Investigators based on the schedule.
II-3 What TAF provides:
- Cryo-preserve minimal 250 embryos at 8-cell stage
- Superovulate juvenile female mice (may train PI's staff to perform the superovulation if PI's protocol permits)
- Instruct PI's staff to mate 15 stud males weekly with superovulated females
- Test and thaw an aliquot of each freezing session to insure the viability of the frozen embryos
Note: On average it takes 2 or 3 embryo collection sessions to freeze down enough embryos to guarantee recovery. However, it may take more sessions if the mouse strain has a low superovulation rate, the stud males have low fertility, or males are too old. Some strains can not be successfully cryopreserved.
III. Freezing mouse embryos at 2-cell stage TAF is implementing cryo-preserve mouse embryos at 2-cell stage by Speed Cryo. Speed Cryo relies on in vitro fertiliztion (IVF) to generate 2-cell embryos for cryopreservation. The advantage of Speed Cryo is that it requires fewer males to generate many embryos from a single IVF procedure. The disadvantage of the Speed Cryo is that it depends on efficiency of mouse IVF, which heavily depends on the strain background and the fertility of the individual males.
